Changes of protein phosphotase-2A level in lymphocytes of type 2 diabetes mellitus accompained by Alzheimer′s disease

Abstract:

Objective

To explore the alterations of protein phosphotase-2A (PP-2A) level in lymphocytes in the patients with type 2 diabetes mellitus (type 2 DM) accompained by Alzheimer′s disease(AD).

Methods

From July 2016 to October 2017, 57 patients in Neurology department of east section, Shanghai Sixth People′s Hospital East and the First People′s Hospital of Zibo were enrolled. All patients were divided into AD group (n=20), type 2 DM (n=20) and type 2 DM accompained by AD group (n=17). Healthy subjects (n=23) of the east section of Shanghai Sixth People′s Hospital for the same period were selected as the control group. The activity of PP-2A was determined by 32P-labelling, and the PP-2A expression of each group was measured by western-blotting.

Results

Compared with control group(0.99±0.06, 1.02±0.09), the activity and the expression of PP-2A were decreased in type 2 DM group (0.87±0.11, 0.91±0.10), AD group(0.76±0.13, 0.86±0.11) and type 2 DM accompained by AD group(0.62±0.72, 0.71±0.09), the differences were statistically significant (F=5.27, 7.74, all P<0.05). Furthermore, the activity and the expression of PP-2A in type 2 DM accompained by AD group were lower than those in type 2 DM group (LSD-t=7.40, 5.85, P<0.05) and AD group (LSD-t=4.56, 4.57, P<0.05). The activity of PP-2A in AD group was lower than that in type 2 DM group (LSD-t=2.84, P<0.05), but the expression of PP-2A between the two groups was not statistically significant (LSD-t=1.28, P>0.05).

Conclusion

Lower activity and expression of PP-2A level in lymphocytes in type 2 DM accompained by AD patients, implying a possible value for diagnosis and treatment.
