Abstract:
Objective To investigate the expression changes of Lumican and autophagy-related proteins in placenta of patients with preeclampsia.
Methods The placental specimens from 19 patients with preeclampsia and 21 healthy puerpera in the Obstetrics Department of People′s Hospital of Rizhao from January to December 2019 were collected. RT-qPCR and Western blotting were used to detect the expression of Lumican in human placenta. The expression levels of autophagy marker proteins microtubule-associated protein light chain 3 (LC3B) and Beclin1 in placental tissues were observed by immunohistochemical staining. The vitality of superoxide dismutase (SOD) and level of malondialdehyde (MDA) in placenta tissues were detected to analyse the level of oxidative stress.
Results The expressions of Lumican mRNA (1.04±0.11) and protein (0.17±0.02) in PE group were lower than those in control group (2.81±0.11, 0.98±0.15, t=25.52, 12.07, all P<0.01). Immunohistochemical results showed that the expressions of Beclin1 and LC3B in PE group were lower than those in control group. The vitality of SOD in PE group [(201.31±26.11)U/mgprot] remarkablely decreased compared with that in control group [(241.65±25.28)U/mgprot] (t=3.50, P<0.01), while the level of MDA in PE group [(6.70±1.10)nmol/mgprot] remarkablely increased compared with that in control group [(5.10±1.05)nmol/mgprot] (t=3.33, P<0.01).
Conclusion There is higher Lumican expression in human placental tissue, but the expression level of Lumican in placental tissue significantly decreases in PE, meanwhile it shows a higher level of oxidative stress, and decreased expression of autophagy related proteins.
