The influence of room temperature and humidity on the measurement results of hepatitis C virus antibody using high-throughput ELISA method

Abstract:

Objective

To explore the influence of room temperature and humidity on the measurement results of hepatitis C virus antibody using high-throughput ELISA method.

Methods

To detect the S/CO values of HCV antibody standard using addcare automatic enzyme immunoassay instrument (ELISA 1100) under 24℃ and different humidity (30%RH, 40%RH, 50%RH, 60%RH, 70%RH, 80%RH, 90%RH) and 50%RH and different temperatures (16℃, 18℃, 20℃, 22℃, 24℃, 26℃, 28℃, 30℃, 32℃, 34℃, 36℃, 38℃, 40℃) respectively.

Results

The S/CO values of HCV-Ab standard showed significant difference under different temperature (16℃, 18℃, 20℃, 22℃, 24℃, 26℃, 28℃, 30℃, 32℃, 34℃, 36℃), and the S/CO value of standard was positively correlated with temperature.The S/CO value altered little between 36℃ and 38℃, but there was of significant difference between 38℃and 40℃.While the S/CO value of the standard gradually decreased with the rising of humidity(30%RH, 40%RH, 50%RH, 60%RH, 70%RH, 80%RH, 90%RH), which presented significant difference between different humidity (P<0.05), and S/CO value of the standard had a negative correlation with the humidity.

Conclusions

Room temperature and humidity have important effects on high-throughput ELISA detection results.High-throughput ELISA should been carried out in the environment with no more than 36℃ and low humidity.Comprehensive consideration is recommended for the automatic enzyme immunoassay instrument with operating temperature range from 18 to 25 degrees, humidity control between 35%RH~50%RH.
